تأثیر تمرینات تداومی و تناوبی شدید به همراه مصرف رزوراترول بر سطوح پروتئین های 3‌SIRT و 1‌OGG بافت کبد موش های نر

چکیده

 هدف پژوهش: سیرتوئین -3 (SIRT3) یک استیل­زدای میتوکندریایی است که از طریق استیلاسیون پروتئین اگزو گوانین گلیکوسیلاز-1 (OGG1)، سبب ترمیم آسیب های DNA میتوکندریایی و جلوگیری از آپوپتوزیس سلولی می گردد. هدف از مطالعه حاضر بررسی تاثیر شش هفته تمرین تداومی و تناوبی شدید همراه با مصرف رزوراترول، بر سطوح SIRT3 وOGG1 بافـت کبد موش های صحرایی نر ویستار بود.روش پژوهش: 48 موش صحرایی بطور تصادفی به شش گروه هشت تایی شامل گروه های کنترل، مکمل، تمرین تداومی، تمرین تداومی + رزوراترول، تمرین تناوبی، و گروه تمرین تناوبی + رزوراترول تقسیم شدند. تمرینات تداومی و تناوبی به مدت شش هفته، شش روز در هفته، به ترتیب با شدت 80 و 95 - 100 درصد  VO2max بر روی نوارگردان مخصوص موش ها اجرا گردید. گروه­های دریافت کننده رزوراترول روزانه 50 میلی­گرم به ازای هر کیلوگرم وزن بدن مکمل دریافت نمودند. پس از مداخله، حیوانات بی هوش و نمونه کبدی آن ها جهت تجزیه و تحلیل جدا گردید.یافته ها: پس از شش هفته در گروه های تمرین تداومی (0001/0=p) و تمرین تداومی + رزوراترول (0001/0=p) سطوح OGG1 کبد افزایش یافت. همچنین شش هفته مصرف رزوراترول (01/0 =p)، تمرین تناوبی (04/0 =p)، تمرین تناوبی + رزوراترول (04/0 =p)، تمرین تداومی (001/0=p)، و تمرین تداومی + رزوراترول (001/0=p) سبب افزایش سطح SIRT3 بافت کبد گردید؛ این در حالی بود که افزایش این دو متغیر در گروه تمرین تداومی + مکمل به طور معنی داری  بیشتر از سایر گروه ها بود (001/0=p).نتیجه گیری: در مقایسه با تمرین تناوبی شدید، اجرای تمرینـات تدوامی به همراه مصرف رزوراترول می تواند بهبودی بیش تری در عملکرد و بیوزنز میتوکندریایی بافت کبد؛ ایجاد کند.

چکیده [English]

Perpose: Sirtuins-3 (SIRT3) is a mitochondrial deacetylase and through acetylation of Oxoguanine Glycosylase 1(OGG1) protein caused repair mitochondrial DNA damage and prevents cell apoptosis. The purpose of this study was to evaluate the effect of vigorous continuous and interval exercise training along with resveratrol on SIRT3 and OGG1 proteins in the liver tissue of male Wistar rats.
Methods: Forty eight male Albino Wistar rats divided into six equal groups including sedentary control, resveratrol, interval exercise, interval exercise + resveratrol, continuous exercise and continuous exercise+ resveratrol. Continuous and interval exercise performed for 6 weeks, six days per week, up to 80% and 95 -100% VO2max respectively on a rat treadmill. Resveratrol users received daily supplements as 50 mg/kg of body weight. After intervention, the animals were anesthetized and their liver samples were taken for analysis.
Results: After 6 weeks of vigorous exercise training and resveratrol consumption, OGG1 level was significantly increased in continuous exercise (p=0.0001) and continuous exercise + resveratrol (p = 0.0001) groups. In addition, the level of liver SIRT3 increased in the resveratrol (p = 0.01), interval exercise (p = 0.04), interval + resveratrol (p = 0.04), continuous exercise (p = 0.001) and continuous exercise + resveratrol (p = 0.001) groups; so that the induced elevation in the dependent variables of continuous + resveratrol group was significantly higher than other groups (p = 0.001).
Conclusion: Compare to the interval exercise, performing of continuous exercise along with resveratrol consumption can induce more improvement in the liver mitochondrial function and biogenesis.
